The highway was closed for the rest of the day, between Silo and Tuppal Rds, with traffic diverted via Silo Rd before reopening early Wednesday morning.
Both drivers, aged in their 50s, were treated at the scene by NSW Ambulance paramedics.
The driver of the car was airlifted to The Alfred hospital in Melbourne in a critical condition.
The driver of the truck was transported by NSW Ambulance to GV Health in Shepparton for treatment.
Police said a tipper truck towing a dual axle trailer collided with a Ford sedan before the sedan then caught on fire.
In providing traffic advice on the incident, Berrigan Shire Council confirmed one of its vehicles had been involved.
It said council officers are working closely with authorities.
Police are appealing for any witnesses to the incident, including anyone that may have dashcam footage.
Emergency calls relating to the crash started about noon last Tuesday, with initial reports of possible persons trapped and vehicles alight.
Two Rural Fire Service NSW trucks were dispatched first, and by 1pm, Fire + Rescue NSW, police and ambulance crews were also on scene.
Back-up support was dispatched from multiple agencies in Deniliquin.
An FRNSW representative said all occupants were able to exit the vehicles, but one patient had been transported to hospital by helicopter.
